Preeclampsia risk prediction from prenatal cell-free DNA screening
- Diagnostic accuracy study
- development and validation cohorts
- external validation cohort
Authors' conclusion
We demonstrate that assessment of cfDNA nucleosome accessibility from early-pregnancy cfDNA sequence data enables the detection of early placental and endothelial-tissue aberrations and may aid in the determination of preeclampsia risk.
Study context
- Population
- Pregnant women with routinely collected clinical prenatal cell-free DNA (cfDNA) screening samples, early pregnancy (≤16 weeks)
- Participants
- 1,854 samples
- Outcome
- Prediction of preterm preeclampsia (AUC)
- Follow-up
- ≤16 weeks' gestation
Performance
As reported in the abstract
Preterm preeclampsia, validation cohort (831 samples)
| Test or model | AUC | Sensitivity at 80% specificity |
|---|---|---|
| cfDNA nucleosome accessibility model | 0.85 | 81% |
Further findings
| Outcome | Values |
|---|---|
| External validation cohort (141 samples) | cfDNA nucleosome accessibility model AUC 0.84 · Sensitivity 79% |
